L-Ergothioneine for Women: A Complete Guide
L-Ergothioneine is becoming one of the most talked-about ingredients in women's health.

Study 1: DR.ERGO® 8-Week RCT
Publication: medRxiv, October 2025
Participants: 66 women aged 35–59
Dosage: 30mg daily, 8 weeks
Result: Measurable improvements across six skin parameters — pigmentation, erythema, glossiness, elasticity, wrinkles, and spots (p<0.01). No adverse events.
Study 2: Mushroom Extract 12-Week RCT
Publication: Frontiers in Medicine, 2024 (Hanayama et al.)
Participants: 80 healthy women
Dosage: 25mg daily via mushroom extract, 12 weeks
Result: Skin hydration improved compared to placebo at the 8-week mark.
Why does oral supplementation reach the skin?
The OCTN1 transporter mentioned earlier is highly expressed on skin keratinocytes and fibroblasts. Once L-Ergothioneine enters the bloodstream, skin cells actively pull it in and deliver it to their mitochondria.
This also explains why L-Ergothioneine has long appeared in premium skincare lines from Clinique, Estée Lauder, La Mer, and Origins.

- AMH increased 38% (from 1.79 ng/mL to 2.47 ng/mL, p<0.05)
- FSH decreased 14.2% (p<0.05)
- Estrogen E2 increased 37.9% (p<0.05)
- Kupperman score (menopausal symptoms) decreased 64.8%
- Sleep quality improved 29.24%

Sleep: A Secondary Observation from the Liver Function Trial
GeneIII's liver function trial (ChiCTR2400093739) was designed to evaluate liver enzyme markers — but also recorded changes in sleep.
60mg daily × 30 days → sleep disturbance scores decreased from baseline (p<0.05).
Separately, a 4-week study (20mg/day) published on MDPI by a Japanese team in 2025 observed similar improvements in sleep-related markers.
Cognition: Ongoing Research
L-Ergothioneine can cross the blood-brain barrier — the protective shield that blocks most external substances from entering the brain. Very few natural antioxidants can actually reach brain tissue, which is one of the most metabolically active organs in the body.

Independent research: A 16-week RCT published in 2025 is evaluating cognitive function in 147 adults aged 55–79 with subjective memory concerns, using 10 or 25mg/day of ErgoActive® (data available on MDPI).
Energy and Longevity: The Relationship with NMN
The longevity supplement getting the most attention right now is NMN (β-nicotinamide mononucleotide). Its job is to raise levels of NAD+ — the "energy currency" of the cell. NAD+ declines with age, affecting energy, metabolism, and how quickly the body repairs itself.

L-Ergothioneine and NMN aren't replacements. They work at different points in the same system:
- NMN supplies the raw material — NAD+ precursors delivered to the cell

A 56-day RCT published in 2025 in Current Developments in Nutrition (the journal of the American Society for Nutrition) studied 62 women aged 36–60 using L-Ergothioneine with PQQ and NR (an NMN relative, also a NAD+ precursor).
The researchers linked all three ingredients to the same cellular energy system.
If you're already taking NMN, L-Ergothioneine doesn't conflict with it — mechanistically, it acts further upstream in the NAD+ pathway.
